The 5-JOET modification incorporates a fluorescent 6-carboxy-4',5'-dichloro-2',7'-dimethoxyfluorescein (JOE) moiety at the 5' terminus of an oligonucleotide. This 5’ JOE modification is conjugated to a modified thymidine which may participate in hybridization. 5' JOE modified oligonucleotides can be used in a wide array of applications, including dual-labeled fluorogenic probes for real-time PCR. JOE is a xanthene dye and this product is prepared using the 6-carboxy isomer. JOE fluoresces in the yellow region of the visible spectrum and can be effectively quenched by BHQ®-1 dye.
