The 5MedC modification incorporates a 5-Methyl deoxyCytidine internally or at the 5' terminus of an oligonucleotide. When substituted for dC, 5-Methyl deoxyCytidine will increase the stability of the duplex, raising the Tm by 1.3°C per insertion. Substitution of dC with 5MedC in the CpG motif of antisense oligonucleotides can prevent or significantly reduce any unwanted pro-inflammatory immune responses in vivo. Other areas of research include the study of DNA methylation in epigenetics.
