The 3-ROX modification adds a fluorescent carboxy-x-rhodamine (ROX) moiety to the 3’ end of an oligonucleotide. 3' ROX labeled oligonucleotides are used in a wide array of applications, including dual-labeled fluorogenic probes for real-time PCR. This 3' ROX modification is conjugated to a modified cytidine which may participate in hybridization. The 3’ ROX modification also introduces a 3' phosphate which prevents extension of the oligonucleotide by DNA polymerase. ROX is a rhodamine dye that fluoresces in the red region of the visible spectrum and can be effectively quenched by BHQ®-2 dye.
