The I-FAM modification incorporates a fluorescent fluorescein (FAM) moiety internally or at the 5' terminus of an oligonucleotide. FAM modified oligonucleotides can be used in a wide array of applications, including dual-labeled fluorogenic probes for real-time PCR. FAM is a xanthene dye and this product is prepared using the 6-carboxy fluorescein isomer. The fluorophore is conjugated to a modified thymidine which may participate in hybridization. FAM fluoresces in the green region of the visible spectrum and its fluorescence can be effectively quenched by BHQ®-1 dye.
